Phase 2b Study Misses Primary Endpoints in Early-Stage Parkinson's Disease

Biogen and Denali Therapeutics have announced topline results from the Phase 2b LUMA clinical trial evaluating BIIB122 (DNL151), an investigational LRRK2 inhibitor for early-stage Parkinson's disease. The study did not achieve its primary or secondary efficacy endpoints, leading both companies to discontinue the development of BIIB122 for idiopathic Parkinson's disease. Denali will, however, continue its independently sponsored Phase 2a BEACON study in patients with Parkinson's disease associated with pathogenic LRRK2 gene variants.

Evaluating a Disease-Modifying Therapy

The global Phase 2b LUMA trial enrolled individuals with early-stage Parkinson's disease to determine whether BIIB122 could slow disease progression by inhibiting the LRRK2 protein, a therapeutic target linked to Parkinson's disease biology. Despite encouraging earlier clinical data, the latest study did not demonstrate meaningful clinical benefit compared with placebo.

Focus Shifts to Genetically Defined Parkinson's Disease

Although the LUMA program has concluded, Denali will continue investigating BIIB122 in the ongoing Phase 2a BEACON study involving patients who carry pathogenic LRRK2 variants. Researchers believe this genetically defined patient population may provide additional insights into the therapeutic potential of LRRK2 inhibition and help guide future precision medicine approaches.
