Cell Signaling Technology (CST) has developed and validated a more environmentally friendly alternative to Triton X-100, a commonly used nonionic surfactant in laboratory workflows. The initiative is part of CST's broader green chemistry efforts to reduce the environmental impact of laboratory chemicals while maintaining scientific performance.

Why Replace Triton X-100?

Triton X-100 has traditionally been used for applications such as cell permeabilization, cell lysis and protein extraction. However, its environmental persistence and degradation products have raised concerns, leading to tighter regulatory requirements and increasing demand for safer laboratory alternatives.

CST Evaluates Safer Surfactants

CST researchers from its Research & Development, Operations, Compliance and Production teams evaluated several alternative detergents. The alternatives were tested for cell permeabilization and lysis performance, antibody compatibility, signal-to-noise ratio, stability across different pH and temperature conditions, and formulation robustness.

ECOSURF SA-9 Selected

Following testing, ECOSURF SA-9 was identified as the preferred alternative for CST's cell-fractionation workflow. The surfactant is derived from seed oil and was selected for its ability to provide similar surfactant properties while supporting the principles of green chemistry and safer degradation.

Maintaining Research Performance

CST's immunofluorescence testing found that several alternatives produced results comparable to or better than Triton X-100 in selected experiments. The company noted that the most suitable detergent can depend on the antibody and application, highlighting the importance of validating alternatives within individual workflows.

The greener detergent is now incorporated into CST's Cell Fractionation Kit (Eco-friendly detergent), #42193, providing researchers with an alternative to Triton X-100 for applicable cell-fractionation workflows.
