Description
- General description: The NELL-1 (neural EGFL like 1) gene encodes an extracellular glycoprotein and is mapped to human chromosome 11p15.[1][2] Nell-1 is a 810 amino acids protein containing thrombospondin-1 module at its N-terminus and includes other motifs : five von Willebrand factor C domains and six epidermal growth factor (EGF)-like domains.[3][2]

- Biochem/physiol Actions: NELL-1 (neural EGFL like 1) is associated with the risk of Rhabdomyosarcomas and the gene is identified to be hypomethylated.[4] The protein encoded influences osteogenic related processes and thus loss of the NELL-1 gene function leads to abnormality in bone and cartilage.[3] NELL-1 is considered as a essential factor in skeletal development as it controls the action of runt-related transcription factor 2.[5] Lack of Nell-1 protein decreases the expression of gene encoding cartilage proteins such as collagens, tenascins, matrilins, and chondroadherin. It serves as an important growth factor in promoting regeneration of articular cartilage.[3] NELL-1 is a tumor suppressor gene and hypermethylation of its promoter region indicates poor prognosis of the disease in esophageal adenocarcinoma.[6]
