- ITGA2B encodes integrin alpha chain 2b
- Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain
- Alpha chain 2b undergoes post-translational cleavage to yield disulfide-linked light and heavy chains that join with beta 3 to form a fibronectin receptor expressed in platelets that plays a crucial role in coagulation
- Mutations that interfere with this role result in thrombasthenia
- In addition to adhesion, integrins are known to participate in cell-surface mediated signalling
- CD41 (platelet glycoprotein IIb, ITGA2B) is composed of two subunits -120 kDa a, alpha and 23 kDa b, beta- that interact with CD61 in the presence of calcium to form a functional adhesive protein receptor
- CD41 is also involved in blood coagulation by mediating platelet aggregation
- Upon blood vessel damage, CD41 binds to a variety of proteins including von Willebrand factor, fibrinogen, fibronectin and vitronectin
- CD41 is mainly expressed on megakaryocyte-platelet lineage, but generally belongs to the antigens that are expressed during the early stages of hematopoietic differentiation
- Diseases associated with CD41 dysfunction include Glansmann Thrombasthenia, and Platelet type-16 bleeding disorder.
