Description

• PD-L1 and PD-L2 are newly discovered members of the B7 family are the ligands for the program death 1 (PD-1) receptor
• They are expressed on immature dendritic cells and mature dendritic cells
• PD-L1 is expressed on antigen-presenting cells, including monocytes stimulated with IFN- γ, and activated human and murine dendritic cells
• Monoclonal antibodies that block PD-L1 and PD-L2 on dendritic cells result in enhance T cell proliferation and cytokine production
• PD-L1 is also expressed on placental trophoblasts, myocardial endothelium, cortical thymic epithelial cells, and on most carcinomas
• Studies show overlapping functions of PD-L1 and PD-L2 and indicate an important role for the PD-L:PD-1 pathway in regulating T cell responses
• The antibody was conjugated to BD Horizon™ BV650 which is part of the BD Horizon Brilliant Violet™ family of dyes
• This dye is a tandem fluorochrome of BD Horizon BV421 with an Ex Max of 405-nm and an acceptor dye with an Em Max at 650-nm
• BD Horizon BV650 can be excited by the violet laser and detected in a filter used to detect APC-like dyes (eg, 660/20-nm filter)
• Due to the excitation and emission characteristics of the acceptor dye, there will be spillover into the APC and Alexa Fluor™ 700 detectors
• However, the spillover can be corrected through compensation as with any other dye combination.
