- M011B targets IL-10 in ELISA and Western blot applications and shows reactivity with human samples
- The M011B immunogen is recombinant human IL-10 (COS cell-derived)
- The original parent clone is 12G8 and has been subcloned as 2553.2.4
- M011B detects IL-10 which has a predicted molecular weight of approximately 19 kDa
- The M011B IL10 antibody (biotinylated conjugate of clone 12G8) has successfully been paired as the detection antibody in a sandwich ELISA with capture antibody M010 (clone 9D7)
- Typical dilutions for sandwich ELISA include 1 μg/mL for coating and 0.125 - 0.25 μg/mL for detection
- Biotinylated antibody M011B (clone 12G8) and antibody M010 (clone 9D7) have successfully been used in combination with recombinant IL10 protein SIL10 in ELISA applications
- This antibody is produced by injecting Rat IgG secreting hybridoma cells into the peritoneum of mice
- The resulting ascites is collected from the mice and the antibody is purified
- IL-10 encodes a protein that acts as a cytokine and is primarily produced by monocytes, with some production by lymphocytes
- This cytokine has various effects on immunoregulation and inflammation, including down-regulating the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages
- It also enhances B cell survival, proliferation, and antibody production
- Additionally, IL-10 can block NF-kappa B activity and is involved in the regulation of the JAK-STAT signaling pathway
- Knockout studies in mice have suggested that this cytokine is an essential immunoregulator in the intestinal tract
- Mutations in this gene have been associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis
- Diseases associated with IL10 include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1.
