- Capsid protein VP1 (UniProt: Q6JC40) is encoded by the Cap gene in Adeno-associated virus 9 (AAV9)
- Adeno-associated viruses (AAV) are small non-enveloped DNA viruses that can serve as promising viral vectors for gene delivery, but can also elicits cellular and humoral immune responses
- AAVs are reported to be nonpathogenic in humans and animals and display low immunogenicity
- Following their attachment to cell surface receptor they are internalized via endocytosis and their intracellular trafficking is a rate-limiting, slow and inefficient process
- AAVs contain an icosahedral capsid, which is 22 to 26 nm in diameter
- They are shown to have a single-stranded DNA genome of approximately 4.7 kilobases that is flanked by two T-shaped inverted terminal repeats (ITRs)
- The 7C7 an epitope-tag antibody recognizes a basic ten amino acid epitope (QPAKKRLNFGQTGD) with high specificity
- The basic nature of the BASE-tag epitope sequence distinguishes it from other commonly used epitope tags, such as Myc, EQKLISEEDL, FLAG, DYKDDDDK, HA, YPYDVPDYA, V5 etc
- Clone 7C7 recognizes capsid proteins VP1 and VP2, but not VP3
