Test Specificity

It recognizes a protein of ∼35kDa, identified as CD74 (Workshop IV). CD74 is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. CD74 is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. Anti-CD74 stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas. Anti-CD74 has been shown to be useful in differentiating atypical fibroxanthoma (-) from malignant fibrous histiocytoma (+).

Antigen:
Bax

Classification:
Monoclonal

Conjugate:
Janelia Fluor 646

Formulation:
50mM Sodium Borate with 0.05% Sodium Azide

Gene Symbols:
BAX

Immunogen:
A synthetic peptide, aa 3-16 (Cys-GSGEQPRGGGPTSS) of human bax protein. (Uniprot: Q07812)

Quantity:
0.1 mL

Primary or Secondary:
Primary

Test Specificity:
Recognizes a protein of 21kDa, identified as the Bax protein. This monoclonal antibody is highly specific to Bax and shows no cross-reaction with Bcl-2 or Bcl-X protein. Bcl-2 blocks cell death following a variety of stimuli. Bax has extensive amino acid homology with Bcl-2 and it homodimerizes and forms heterodimers with Bcl-2. Overexpression of Bax accelerates apoptotic death induced by cytokine deprivation in an IL-3 dependent cell line, and Bax also counters the death repressor activity of Bcl-2.

Antigen:
CD45RO

Classification:
Monoclonal

Conjugate:
APC-Cyanine7

Formulation:
PBS with 0.05% Sodium Azide

Gene Symbols:
PTPRC

Immunogen:
Interleukin-2-dependent human T lymphocytes were used as the immunogen for the CD45RO antibody.

Quantity:
0.1 mL

Primary or Secondary:
Primary

Test Specificity:
Recognizes a 180-185kDa protein, identified as isoform of leukocyte common antigen (CD45RO) (4th Leucocyte Typing Workshop: Code No. N31). The epitope recognized by this antibody is sensitive to neuraminidase digestion. This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. It rarely stains NK cells or B-cell lymphomas.
