Test Specificity

This monoclonal antibody reacts with a CD32 (FcgRII) epitope distinct from that defined by monoclonal antibody 8.26 and the epitope overlaps with that of monoclonal antibody 7.30 (cluster 4). It displays a stronger reaction with Daudi than with U937 cells. The epitope is located in domain 2 of FcgRIIa. Its Fab'2 fragments block immune complex binding. CD32 (FcRII) is a type 1 transmembrane glycoprotein that mediates several functions including phagocytosis, cytotoxicity, and immunomodulation as well as platelet aggregation. Three genes (A, B, and C) encode CD32 and at least 6 isoforms are generated via alternative mRNA splicing, i.e., IIa1, IIa2, IIb1, IIb2, IIb3 and IIc. Monocytes/macrophages, placental trophoblasts and endothelial cells express all isoforms. In addition, the IIb isoform is expressed by B cells, and the IIa isoform by platelets, granulocytes and, weakly, by B cells. NK cells and neutrophils express Isoform IIc. CD32 binds weakly to the Fc region of monomeric IgG but m

Antigen:
MUC1

Classification:
Monoclonal

Conjugate:
APC-Cyanine7

Formulation:
PBS with 0.05% Sodium Azide

Gene Symbols:
MUC1

Immunogen:
Human breast cancer cell line ZR-75 cells

Quantity:
0.1 mL

Primary or Secondary:
Primary

Test Specificity:
This monoclonal antibody recognizes full-length MUC1 in a glycosylation-independent manner and can bind to the fully glycosylated protein. The dominant epitope of this monoclonal antibody is APDTR in the VNTR region. It reacts with the core peptide of the MUC1 protein, which is a member of a family of mucin glycoproteins that are characterized by high carbohydrate content, O-linked oligosaccharides, high molecular weight (200kDa) and an amino acid composition rich in serine, threonine, proline and glycine. The core protein contains a domain of 20 amino-acid tandem repeats that functions as multiple epitopes for the monoclonal antibody. Incomplete glycosylation of some tumor-associated mucins may lead to variable unmasking of the multiple peptide epitopes leading to the observed differences in staining intensity between normal and malignant tissues. This monoclonal antibody reacts with both normal and malignant epithelia of various tissues including breast and colon.
