Test Specificity

This MAb reacts with a protein of 20-30kDa, identified as PGP9.5, also known as ubiquitin carboxyl-terminal hydrolase-1 (UchL1). Initially, PGP9.5 expression in normal tissues was reported in neurons and neuroendocrine cells but later it was found in distal renal tubular epithelium, spermatogonia, Leydig cells, oocytes, melanocytes, prostatic secretory epithelium, ejaculatory duct cells, epididymis, mammary epithelial cells, Merkel cells, and dermal fibroblasts. Furthermore, immunostaining for PGP9.5 has been shown in a wide variety of mesenchymal neoplasms as well. A mutation in PGP9.5 gene is believed to cause a form of Parkinson's disease.

Test Specificity:
This monoclonal antibody reacts with an extracellular domain (close to transmembrane) of CD56/NCAM. Three isoforms of neural cell adhesion molecule (NCAM) are produced by differential splicing of the RNA transcript from a single gene. The 135kDa isoform is the basic molecule, which is glycosylated or sialylated to produce the mature species. Anti-CD56 recognizes two proteins of the neural cell adhesion molecule, the basic molecule expressed on most neuroectodermally derived tissues and neoplasms (e.g. retinoblastoma, medulloblastomas, astrocytomas, neuroblastomas, and small cell carcinomas). It is also expressed on some mesodermally derived tumors (rhabdomyosarcoma). Anti-CD56 plays an important role in the diagnosis of nodal and nasal NK/T-cell lymphomas.

Test Specificity:
This monoclonal antibody reacts with the rod domain of human cytokeratin-19 (CK19), a polypeptide of 40kDa. Its epitope maps between amino acid 312-335. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Recognizes a protein of 36kDa, identified as cyclin D1. Cyclin D1, one of the key cell cycle regulators, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms. This antibody neutralizes the activity of cyclin D1 in vivo. About 60% of mantle cell lymphomas (MCL) contain a t(11; 14)(q13; q32) translocation resulting in over-expression of cyclin D1. This antibody is useful in identifying mantle cell lymphomas (cyclin D1 positive) from CLL/SLL and follicular lymphomas (cyclin D1 negative). About 40% of breast carcinomas are positive for Cyclin D1. Occasionally, hairy cell leukemia and plasma cell myeloma weakly express Cyclin D1.
