This antibody recognizes actin of skeletal, cardiac, and smooth muscle cells. It is not reactive with other mesenchymal cells except for myoepithelium. Actin can be resolved on the basis of its isoelectric points into three distinctive components: alpha, beta, and gamma in order of increasing isoelectric point. Anti-muscle specific actin recognizes alpha and gamma isotypes of all muscle groups. Non-muscle cells such as vascular endothelial cells and connective tissues are non-reactive. Also, neoplastic cells of non-muscle-derived tissue such as carcinomas, melanomas, and lymphomas are negative. It stains tumors of smooth muscle (leiomyomas and leiomyosarcomas) as well as skeletal muscle (rhabdomyomas and rhabdomyosarcomas).

Antigen:
Vimentin

Classification:
Monoclonal

Conjugate:
Unconjugated

Formulation:
10mM PBS and 0.05% BSA with 0.05% Sodium Azide

Gene Alias:
FLJ36605, vimentin

Host Species:
Mouse

Purification Method:
Protein L or PEG purified

Regulatory Status:
RUO

Primary or Secondary:
Primary

Test Specificity:
This monoclonal antibody reacts with a 58kDa protein identified as vimentin. It reacts with a non-hematopoietic epitope of vimentin and shows no cross-reaction with other closely related intermediate filament proteins (IFPs) such as desmin, keratin, neurofilament, and glial fibrillary acid protein. Vimentin is ubiquitously expressed in mesenchymal cells such as fibroblasts, smooth muscle cells, and endothelium. Antibody against vimentin is useful as part of an antibody panel for differential diagnosis of tumors of unknown origin.Ab-2 does not react with leukocyte common antigen-positive tissues such as lymphomas and leukemias.

Expression of Vimentin, when used in conjunction with anti-keratin, is helpful when distinguishing melanomas from undifferentiated carcinomas and large cell lymphomas. All melanomas and Schwannomas react strongly with anti-Vimentin. It labels a variety of mesenchymal cells, including melanocytes, lymphocytes, endothelial cells, and fibroblasts. Non-reactivity of anti-Vimentin is often considered more useful than its positive reactivity, since there are a few tumors that do not contain Vimentin, e.g. hepatoma and seminoma. Anti-Vimentin is also useful as a tissue process control reagent.

Antigen:
NCAM-1/CD56

Classification:
Monoclonal

Conjugate:
Unconjugated

Formulation:
10mM PBS and 0.05% BSA with 0.05% Sodium Azide

Gene Alias:
CD56, CD56 NCAM-1, CD56 antigen, MSK39, N-CAM-1, NCAM-1, NCAMantigen recognized by monoclonal 5.1H11, neural cell adhesion molecule 1, neural cell adhesion molecule, NCAM

Host Species:
Mouse

Purification Method:
Protein A or G purified

Regulatory Status:
RUO

Primary or Secondary:
Primary

Test Specificity:
This monoclonal antibody reacts with an extracellular domain (close to transmembrane) of CD56/NCAM. Three isoforms of neural cell adhesion molecule (NCAM) are produced by differential splicing of the RNA transcript from a single gene. The 135kDa isoform is the basic molecule, which is glycosylated or sialylated to produce the mature species. Anti-CD56 recognizes two proteins of the neural cell adhesion molecule, the basic molecule expressed on most neuroectodermally derived tissues and neoplasms (e.g. retinoblastoma, medulloblastomas, astrocytomas, neuroblastomas, and small cell carcinomas). It is also expressed on some mesodermally derived tumors (rhabdomyosarcoma). Anti-CD56 plays an important role in the diagnosis of nodal and nasal NK/T-cell lymphomas.
