Filaggrin is an intermediate filament-associated protein that aggregates keratin intermediate filaments in mammalian epidermis. It is initially synthesized as a polyprotein precursor, profilaggrin (consisting of multiple filaggrin units of 324 aa each), which is localized in keratohyalin granules, and is subsequently proteolytically processed into individual functional filaggrin molecules. Active filaggrin is present at a level of the epidermis where keratinocytes are in transition between the live nucleated granular layer and the anucleate cornified layer, suggesting that filaggrin aids in the terminal differentiation process by facilitating apoptotic machinery.

This MAb reacts with a protein of 20-30 kDa, identified as PGP9.5, also known as ubiquitin carboxyl-terminal hydrolase-1 (UCH-L1). Initially, PGP9.5 expression in normal tissues was reported in neurons and neuroendocrine cells but later it was found in distal renal tubular epithelium, spermatogonia, Leydig cells, oocytes, melanocytes, prostatic secretory epithelium, ejaculatory duct cells, epididymis, mammary epithelial cells, Merkel cells, and dermal fibroblasts. Furthermore, immunostaining for PGP9.5 has been shown in a wide variety of mesenchymal neoplasms as well. A mutation in PGP9.5 gene is believed to cause a form of Parkinson's disease.

GP2 (glycoprotein 2), also known as ZAP75, is a 537 amino acid secreted protein. It is an integral membrane protein that is secreted from intracellular zymogen granules and associates with the plasma membrane via glycosylphosphatidylinositol (GPI) linkage. GP2 is cleaved and then released into the pancreatic duct along with exocrine secretions. GP2 binds pathogens such as enterobacteria, thereby playing an important role in the innate immune response.
