Test Specificity

Epitope of this monoclonal antibody is located in the 8th type III repeat in the cell-binding region of fibronectin. Fibronectins are disulfide-linked, dimeric glycoproteins of ∼440kDa. They possess at least four binding sites for collagen, glycosaminoglycans, transglutaminase, and a cell surface receptor. Fibronectins are involved in cell adhesion, tissue organization, and wound healing. Fibronectins are present in basement membranes, interstitial connective tissue matrix, and blood. Cellular fibronectin is widely distributed in the stroma of many malignant tumors. This monoclonal antibody is excellent for staining of formalin-fixed, paraffin-embedded tissues.

Antigen:
alpha-Fetoprotein/AFP

Classification:
Monoclonal

Conjugate:
Alexa Fluor 350

Formulation:
50 mM sodium borate with 0.05% sodium azide

Gene Symbols:
AFP

Immunogen:
Human Alpha feto protein (Uniprot: P02771)

Primary or Secondary:
Primary

Test Specificity:
It recognizes an oncofetal glycoprotein with a single chain of 70kDa, which is identified as alpha fetoprotein (AFP). This monoclonal antibody is highly specific to AFP and shows no cross-reaction with other oncofetal antigens or serum albumin. The yolk sac and the liver produce AFP during fetal life. AFP expression in adults is often associated with hepatoma or teratoma. However, hereditary persistence of alpha-fetoprotein may also be found in individuals with no obvious pathology. The protein is thought to be the fetal counterpart of serum albumin, and the AFP and albumin genes are present in tandem in the same transcriptional orientation on chromosome 4. AFP is found in monomeric as well as dimeric and trimeric forms, and binds copper, nickel, fatty acids and bilirubin. The level of AFP in amniotic fluid is used to measure renal loss of protein to screen for spinal bifida and anencephaly.

IGF1

Immunogen:
Recombinant full-length human IGF-I/IGF-1 protein (Uniprot: P05019)

Primary or Secondary:

Test Specificity:
This antibody is specific to Insulin-like Growth Factor (IGF-1) and shows minimal cross-reaction with IGF-11, Proinsulin, MSF, and Insulin. IGF-1 is a polypeptide growth factor with two isoforms that are produced by alternative splicing. Isoform 1 is also known as IGF-IB while isoform 2 is known as IGF-IA. IGF-1 stimulates the proliferation of a wide range of cell types including muscle, bone and cartilage tissue. It functions as an autocrine regulator of growth. Activation of IGF system has emerged as a key factor for tumor progression and resistance to apoptosis in many cancers like those of breast, thyroid and colon.
