  • PGUA is a colorless compound that is hydrolyzed by β-glucuronidase to form nitrophenyl, a yellow compound
  • In the indole test, tryptophanase enzyme attacks the side chain of tryptophan and leaves an aromatic ring in the form of indole
  • This reaction occurs as a condensation process that is formed by an acid splitting of the protein molecule
  • The ρ-dimethylaminobenzaldehyde in Kovacs reagent then combines with indole to form a red color complex 5,6
