Description

  • Identify Enterobacteriaceae on the basis of double sugar fermentation and hydrogen sulphide production using Thermo Scientific™ Oxoid™ Kligler Iron Agar (Dehydrated)
  • Klingler Iron Agar combines the features of Kligler's Lead Acetate medium 1,2,3 and Russell's Double Sugar Agar 4
  • The medium is recommended for the identification of colonies picked off from plating media such as MacConkey Agar, Bismuth Sulphite Agar, or Desoxycholate Citrate Agar, etc
  • Differentiate Enterobacteriaceae on the basis of double sugar fermentation and hydrogen sulphide production with Klinger Iron Agar
  • Non lactose fermenters initially produce a yellow slant and butt as a result of glucose fermentation
  • Once the glucose is depleted, the reactions reverts to red slants (alkaline) due to oxidation of acids
  • Lactose fermenting organisms produce yellow slants and butts
  • There is no reversion to red in the slant because enough acid is produced to maintain an acid pH under aerobic conditions
  • Non-fermenters produce red slants and butts
  • H 2 S production results in a blackening of the medium, either throughout the butt or in a ring formation near the top of the butt
  • Gas production is demonstrated by the presence of bubbles or cracks in the medium
  • Typical reactions observed on Kligler Iron Agar after 18-24 hours at 35 °C
