Description

  • Detect and differentiate Pseudomonas aeruginosa from other Pseudomonas spp
  • using Thermo Scientific™ Remel™ Pseudomonas P Agar (Dehydrated)
  • In 1954, King et al
  • developed two media formulations (Medium A and Medium B) to enhance pigment production in pseudomonads 1
  • Earlier work had demonstrated that the production of pyocyanin and fluorescein is influenced by amino acids, inorganic ions, minerals, and peptones
  • King determined that the composition of the peptone in the medium affected the production of these pigments
  • Pseudomonas aeruginosa is the only species of Pseudomonas known to produce and excrete the pigment pyocyanin
  • Pseudomonas P Agar was a medium developed by King, Ward, and Raney to improve pyocyanin production by Pseudomonas aeruginosa 1
  • The absence of minimal concentrations of components which may have a detrimental effect influences pigment production
  • A combination of magnesium chloride and potassium sulfate was found to be most effective in stimulating pyocyanin production 2
  • The medium contains gelatin and meat peptones that provide nutrients necessary for bacterial growth
  • These peptones contain less than 0.1% phosphorus, which has been found to increase pyocyanin production
  • The total concentration of magnesium chloride and potassium sulfate does not exceed 2%, which stimulates pyocyanin production
  • Addition of glycerol to the prepared medium acts as an energy source.
