Detect the deoxyribonuclease (DNase) activity in microorganisms with Thermo Scientific™ Remel™ DNase Test Agar w/Methyl Green
In 1957, Jeffries et al
developed a medium to demonstrate DNase activity of microorganism that possess this extracellular enzyme 1
This medium is used to identify a variety of bacteria including staphylococci, enteric gram-negative bacilli and pseudomonads 2-5
On DNase Test Agar w/Methyl Green, the colony is surrounded by a clear zone in an otherwise green medium 6
DNase Test Agar w/Methyl Green is a solid medium used to detect the deoxyribonuclease (DNase) activity in microorganisms
For the methyl green modification, the dye methyl green is able to bind to polymerized DNA to form a stable green complex at pH 7.3
The action of DNase depolymerizes the DNA structure releasing methyl green; when not combined at this pH methyl green becomes colorless resulting in colorless halos around DNase-positive colonies
Easy- to-interpret: Positive test – Growth line surrounded by a colorless zone in an otherwise green agar plate
Negative test – Growth line surrounded by no zone, the entire plate is green
Ready- to-use: convenience of prepared media
DNase Test Agar w/Methyl Green contain casein and soy peptone that supply nitrogen, amino acids and peptides necessary for bacterial growth
Sodium chloride provide essential electrolyte which maintain osmotic equilibrium
