  • In 1958, Middlebrook and Cohn first described Middlebrook 7H9 Broth for the cultivation of mycobacteria 1
  • The supplemental nutrients glycerol, oleic acid, albumin, and dextrose were found to enhance the growtth of most mycobacteria 2,3
  • This medium is used as a basal medium to prepare inocula for biochemical identification tests and antibiotic susceptibility testing procedures 4,5
  • Use Middlebrook 7H9 Broth with Glycerol for isolation and cultivation of Mycobacterium species
  • Recommended for identification and susceptibility testing of Mycobacteria , Nocardiae and other aerobic Actinomycetes by CLSI 5

  • Middlebrook 7H9 Broth with Glycerol contains inorganic compounds that supply potassium, sulfur, magnesium, and phosphorus necessary for the growth of mycobacteria
  • Inorganic copper, iron, zinc, and calcium stimulate growth
  • Sodium citrate converts to citric acid in solution and holds certain inorganic cations
  • Ammonium sulfate is a source of nitrogen, dextrose and glycerol provide carbon and albumin binds to the free fatty acids which may be toxic to Mycobacterium species
  • Catalase destroys toxic peroxides and catalyzes the reaction of iron with molecular oxygen
  • Biotin is required for carboxylation and decarboxylation reactions and pyridoxine is a precursor of enzymatic activity.
