Description

• General description: CD27 antigen (UniProt P41272; also known as CD27, CD27L receptor, T-cell activation antigen CD27, Tumor necrosis factor receptor superfamily member 7) is encoded by the Cd27 (also known as Tnfrsf7) gene (Gene ID 21940) in murine species. CD27 is a TNF receptor superfamily member that binds to CD70 and mediates the survival, function, and differentiation of T-, B-, natural killer (NK), and plasma cells. Interaction with CD70 provides costimulation in T cells induces signaling events essential for cell proliferation, long-term maintenance of antigen-specific T cells, antiviral responses, antitumor immunity, and alloreactivity. CD27 ligation on B-cells by CD70 causes enhanced plasma cell formation and increased IgG production. CD27 is also used as a marker for memory B cells in the classification of B-cell deficiencies, including common variable immunodeficiency (CVID). CD27 is synthesized with a signal peptide (a.a. 1-23), which was removed posttranslationally to yield the mature type I transmembrane protein with an extracellular (a.a. 24-182) composed of three TNFR-Cys repeats (a.a. 26-63, 64-104, 105-141), followed by a transmembrane region (a.a. 183-203), and a cytoplasmic tail (a.a. 204-250).
• Specificity: Clone RM27-3E5 enhanced anti-CD3-stimulated T-cell proliferation when immobilized in vitro and augmented OVA-specific CTL induction when administrated in vivo (Roberts, D.J., et al. (2010). J. Immunother. 33(8): 769 779; Sumi, T., et al. (2008). Immunol. Lett. 119(1-2):91-96).

• Application: Neurtralizing Analysis: A representative lot, when injected intraperitoneally following OVA-expressing B16 melanoma cells (B16-cOVA) implant, suppressed B16-cOVA lung metastasis and tumor growth among wild-type, but not Cd27-knockout, mice (Roberts, D.J., et al. (2010). J. Immunother. 33(8): 769 779).Neurtralizing Analysis: A representative lot, when injected intraperitoneally during the induction phase of ragweed-induced experimental allergic conjunctivitis (EC), significantly affected the systemic immune responses, but not the severity of EC in mice (Sumi, T., et al. (2008). Immunol. Lett. 119(1-2):91-96).
• Quality: Evaluated by Flow Cytometry in mouse splenocytes.Flow Cytometry Analysis: 2 µL of this antibody detected CD27-positive mouse splenocytes.
