• General description: Tight junction protein 1/Zona occludens 1 protein (TJP1, ZO-1) is a transmembrane plaque protein belonging to the membrane-associated guanylate kinase (MAGUK) family of scaffold proteins. It consists of three PDZ (PSD-95/Discs large/ZO-1) domains, one SH3 (Src-homology 3) domain, and one GK (guanylate kinase) domain. Additionaly, it also contains a long proline-rich carboxyl terminal region and a ZU5 domain.

• Biochem/physiol Actions: Tight junction protein ZO-1 is a protein encoded by the TJP1 gene in humans and is mapped to chromosome 15q13. It is a membrane-scaffolding protein that plays an important role in maintaining tight-junction integrity, which is disrupted in many invasive cancers and intestinal diseases. It may act as a tumor-suppressor gene in GIST. It is an integral component of functional TJs and is regulated by p38δ. It shows a hypermethylation status in children with non-Hodgkin lymphoma (NHL) and the methylation level is positively correlated with the number of malignant cells in bone marrow. It may be useful as a novel molecular marker in early diagnosis, outcome assessment, prognostic evaluation and detection of minimal residual disease.
