• General description: Formin-like 2 (FMNL2) belongs to the family of diaphanous-related formins (DRF), which are ubiquitously expressed and have highly conserved domains. FMNL2 is highly expressed in mammary and gastrointestinal epithelia, placenta, reproductive tract and lymphatic tissues. FMNL2 has two isoforms due to alternative splicing, and they differ at their C-terminals. FMNL2A and FMNL2B have the characteristic FDD and formin homology1 (FH1) and FH2 domains. This gene is localized to chromosome 2q23.3.

• Biochem/physiol Actions: Formin-like 2 (FMNL2) regulates actin cytoskeleton, and therefore controls cell motility and migration, cell adhesion, cell polarity, filopodium formation and cytokinesis. It mediates the polymerization of actin via its FH2 domain, while its FH1 domain binds to profilin. Cellular morphological changes are induced by FMNL2, via the N-myristoylation of the involved proteins. It also acts as the effector protein for GTPases belonging to Rho signaling pathway. FMNL2 is down-regulated in hepatocellular carcinoma as compared to normal hepatic epithelial cells. The lower expression of FMNL2 predicts poor prognosis and survival for hepatocellular carcinoma patients. In colorectal carcinoma, it has been implicated in maintaining the epithelial-mesenchymal transition (EMT). Studies show that this protein plays a key role in the metastasis of colorectal cancer.
