• General description: TRAF2 (TNF receptor associated factor 2) is one of the six members of the TRAF family. It is an adaptor protein, and is composed of a conserved TRAF domain in its C-terminal and a RING finger domain in its N-terminal. It also contains multiple zinc finger regions.

• Biochem/physiol Actions: TRAF2 (TNF receptor associated factor 2) is recruited on activation of TNFR (tumor necrosis factor receptor) by TNF. It directly binds to TNFR2 and interacts with TNFR1 through TRADD (TNFR1-associated death domain protein). This protein activates JNK (c-jun N-terminal kinase) upon activation by ASK1 (apoptosis signal-regulating kinase 1). TRAF2 faciliates rapid vaccinia virus (VACV) entry and thus aids VACV replication. TRAF2 and RIP1 (receptor-interacting protein) up-regulation in leukocytes results in increased inflammatory responses in multiple sclerosis (MS) patients, and the elevation of this protein is seen in patients with relapsing-remitting multiple sclerosis (RRMS). The expression of this protein is linked with poor prognosis in activated B-cell (ABC)-like diffuse large B-cell lymphomas (DLBCL), where it activates NF-κB. It might also be the reason for chemoresistance in ABC-DLBCL.
