• General description: CUB-domain-containing protein 1 (CDCP1) is a type I transmembrane protein which functions as a substrate for Src kinases. It has a molecular weight of 140kDa, and has a proteolytically cleaved form with a molecular weight of 85kDa. It has an intracellular region made of five tyrosines, and a large extracellular region containing the CUB (C1r/C1s, Uegf, Bmp1) domain. Both the isoforms of CDCP1 are expressed in varying ratios in epithelial cells. This gene is localized to human chromosome 3p21.31. CDCP1 protein is composed of 836 amino acids, with a signal peptide of 29 amino acids, and a cytoplasmic domain consisting of 150 amino acids.

• Biochem/physiol Actions: CUB domain-containing protein 1 (CDCP1) is phosphorylated by Src kinases and mediates resistance to anoikis, promotes degradation of extracellular matrix, tumor invasion and metastasis. It is a cancer associated gene and is up-regulated in multiple cancers such as, lung, pancreas and kidney. Its overexpression in these cancers is related to poor prognosis. CDCP1 inhibits integrin-mediated cell adhesion by acting as a substrate of Src-kinases. It also facilitates the metastasis of melanoma, and is associated with poor prognosis of renal cell carcinoma. CDCP1 aids bone morphogenetic protein 4 (BMP4) in initiating epithelial-mesenchymal transition, and suppresses the epithelial phenotype of pancreatic cancer cells. It confers anoikis resistance to lung cancer cells by participating in protein kinase Cδ (PKCδ) signaling.
