Description

• General description: The gene cathepsin B (CTSB) has been mapped to human chromosome 8p22. The gene encodes a lysosomal cysteine protease composed of a dimer of disulphide linked heavy and light chains. Under physiological conditions the protein is localized in lysosomes and in presence of some cellular signals it is released from the lysosomes and trigger different physiological events.[1]

• Application: Anti-CTSB antibody produced in rabbit, a Prestige Antibody, is developed and validated by the Human Protein Atlas (HPA) project . • Biochem/physiol Actions: Cathepsin B (CTSB) has been linked to various responses including lysosomal protein turnover, apoptosis and immune responses. The protein is involved in activation of epithelial Na+ channel in cystic fibrosis lung disease airways. Presence of cocaine along with HIV (human immunodeficiency virus)-1 infection causes increase in CTSB secretion and neurotoxicity. CTSB is up-regulated in synovial fluid of patients suffering with rheumatoid arthritis. The protein is considered as a biomarker for many cancers. Stearoyl-CoA desaturase-5 reduces melanoma malignancy by regulating secretion of CTSB. Mutations in CTSB (C76G/A4383C) have been linked to high risk of hepatocellular carcinoma. CTSB levels and activity were shown to be increased in human esophageal squamous cell carcinoma, breast cancer, colorectal carcinomas and no small cell lung carcinoma.[1][2] CTSB increase the localization of cytoplasmic c-Jun-NH2-terminus kinase, thereby inducing migration of glioma cells and making them more malignant.
