• General description: The gene TNFRSF10B (TNF receptor superfamily member 10b) is mapped to human chromosome 8 and contains 10 exons. It encodes a protein that is commonly referred to as death receptor 5 (DR5) and is a member of tumor necrosis factor receptor (TNFR) superfamily. It contains a characteristic cytoplasmic death domain (DD).

• Biochem/physiol Actions: The gene TNFRSF10B (TNF receptor superfamily member 10b) is a tumor suppressor gene, which upon mutation, leads to a loss of chromosome p arm, a common occurrence in head and neck tumors. It inhibits tumorigenesis via apoptosis. The cytoplasmic death domain (DD) of the protein employs fas-associated death domain (FADD) and caspases to form the death-inducing signal complex (DISC) when the receptor is trimerized. This leads to activation of caspases and cell death.
