• Biochem/physiol Actions: The gene TTI2 (TELO2 interacting protein 2) encodes a protein that forms a conserved trimeric complex called the Triple T complex by associating with telomere maintenance 2 (TEL2) and Tel2-interacting proteins 1 (TTI1). This complex is involved in the regulation of the amount of phosphoinositide-3-kinase-related protein kinase (PIKKs) and DNA damage response signaling. This complex functions in the development and functioning of the human brain. Mutations in TTI2 gene have been associated with reduced amounts of all Triple T complex components and cause severe cognitive impairment, microcephaly, behavioral troubles, short stature, skeletal anomalies, and facial dysmorphic features.
