• Specificity: Clone 3F7 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects JNK1/2 phosphorylated on threonine 183 and tyrosine 185.
• Immunogen: KLH-conjugated linear peptide corresponding to 10 amino acids surrounding phosphothreonine 183 and phosphotyrosine 185 from human JNK/SAPK.
• Application: Anti-phospho JNK (Thr183, Tyr185), clone 3F7 ZooMAb , Cat. No. ZRB1173, is a highly specific recombinant rabbit monoclonal antibody that detects JNK/SAPK phosphorylated on Thr183 and Tyr185 and has been tested for use in Immunocytochemistry, Immunohistochemistry, Peptide Inhibition Assay, and Western Blotting.
• Target description: Mitogen-activated protein kinase 8/9 (UniProt: P45983/P45984; also known as MAP kinase 8/9, MAPK8/9, Stress-activated protein kinase 1c/1a, SAPK1c/SAPK1a, Stress-activated protein kinase JNK1/ JNK2, c-Jun N-terminal kinase 1/2) is encoded by the MAPK8/9 (also known as JNK1/JNK2, PRKM8/9, SAPK1/2, SAPK1C/1A) gene (Gene ID: 5599/ 5601) in human. SAPK/JNK is a serine/threonine-protein kinase that is involved in various biological processes, including cell proliferation, differentiation, migration, transformation, and programmed cell death. It is activated in response to extracellular stimuli such as proinflammatory cytokines or physical stress. Both JNK1 and JNK2 contain a TXY motif (aa 183-185) that contains the threonine and tyrosine residues whose phosphorylation activates these kinases. Protein kinase domain of SAPK/JNK is localized to amino acids 26-321. Phosphorylation and activation of SAPK/JNK is achieved by two dual specificity kinases, MAP2K7 and MAP2K4. Following their activation SAPK/JNK phosphorylate several transcription factors, primarily components of AP-1 such as JUN, JDP2, and ATF2 and thus regulates AP-1 transcriptional activity. It promotes apoptotic cell death in stressed cells by phosphorylating key regulatory factors including p53/TP53 and Yes-associates protein YAP1. The immunogen sequence used to generate this clone is conserved in both JNK1 and JNK2. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
• Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose. Normal appearance is a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
• Reconstitution: 300 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
• Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
