  • Specificity: Clone 4B7 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Connector enhancer of kinase suppressor of ras 1 (CNKSR1). It targets an epitope within 21 amino acids from the C-terminal region. It detects both isoforms of CNKSR1.
  • Immunogen: KLH-conjugated linear peptide corresponding to 21 amino acids from the C-terminal region of human Connector enhancer of kinase suppressor of ras 1 (CNKSR1).

  • Target description: Connector enhancer of kinase suppressor of ras 1 (UniProt: Q969H4; also known as Connector enhancer of KSR 1, CNK homolog protein 1, CNK1, hCNK1, Connector enhancer of KSR-like, CNKSR1) is encoded by the CNKSR1 (also known as CNK1) gene (Gene ID: 10256) in human. CNKSR1 is a peripheral membrane protein that serves as an adapter protein and regulates Ras signaling pathways through interactions with proteins of distinct pathways in the cytoplasm. One of its main function is to regulate cancer-related signaling pathways and integrate output to different intracellular signaling cascades upon stimulation by extracellular cues. Its activity is closely regulated by phosphorylation at tyrosines 26, 519, and 665. Phosphorylation of Tyr 519 is shown to recruit CNKSR1 to the nucleus and additional phosphorylation of tyrosine 26 enables it to promote SRE-dependent gene expression. Overexpression of CNKSR1 has been reported in pancreatic cancer tissue and it is suggested to be an independent prognostic marker of overall survival. At its N-terminal it has a sterile motif (SAM; aa 7-70), followed by a conserved region in Cnk (CRIC; aa 78-164), a), a PDZ domain (aa 196-285), and a pleckstrin homology (PH) domain (aa 403-502). Two isoforms of CNKSR1 have been described that are produced by alternative splicing. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
  • Reconstitution: 30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
