  • Target description: Forkhead box protein O3 (UniProt: O43524; also known as AF6q21 protein, Forkhead in rhabdomyosarcoma-like 1) is encoded by the FOXO3 (also known as FKHRL1, FOXO3A) gene (Gene ID: 2309) in human. Forkhead box protein O3 is a transcriptional activator that triggers apoptosis in the absence of survival factors, including neuronal cell death upon oxidative stress. It recognizes and binds to the DNA sequence 5′-[AG]TAAA[TC]A-3′. Its DNA binding domain located in amino acids 157-251. The nuclear localization signal is localized in amino acids 242-259. In the cytoplasm it is present in an inactive state and it translocates to the nucleus upon oxidative stress and in the absence of survival factors. In response to metabolic stress, it translocates into the mitochondria where it promotes mtDNA transcription. In the presence of survival factors such as IGF-1, it is phosphorylated on threonine 32 and serine 253 by AKT1/PKB. The phosphorylated form then interacts with 14-3-3 proteins and is retained in the cytoplasm. Survival factor withdrawal induces dephosphorylation and promotes its translocation to the nucleus where the dephosphorylated protein induces transcription of target genes and triggers apoptosis. FOXO3 deacetylation by SIRT1 or SIRT2 stimulates its interaction SKP2 and facilitates SCF(SKP2)-mediated FOXO3 ubiquitination and proteasomal degradation. A chromosomal aberration involving FOXO3 is found in secondary acute leukemias. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
  • Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
