Description

  • General description: We are committed to bringing you greener alternative products, which adhere to one or more of The 12 Principles of Green Chemistry. This antibody is Preservative-free, produced without the harm or sacrifice of animals and exceptionally stable to allow for ambient shipping and storage if needed and thus aligns with "Waste Prevention", "Designing Safer Chemicals" and "Design for Energy Efficiency". Click here for more information.
  • Specificity: Clone 1J7 is a ZooMAb® Rabbit recombinant monoclonal antibody that detects MEK2. It targets an epitope within 21 amino acids the N-terminal region. Also, displays reactivity with MEK1.
  • Immunogen: KLH-conjugated linear peptide corresponding to 21 amino acids from the N-terminal region of human MEK2.
  • Application: Quality Control TestingEvaluated by Western Blotting in HeLa cell lysate.Western Blotting Analysis: A 1:1,000 dilution of this antibody detected MEK2 in HeLa cell lysate.Tested applicationsWestern Blotting Analysis: A 1:1,000 dilution from representative lot detected MEK2 in PC12, NIH3T3, and A431 cell lysates.Immunocytochemistry Analysis: A 1:100 dilution from representative lot detected MEK2 in HeLa, A431, HepG2, and NIH 3T3 cells.Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from representative lot detected MEK2 in human spleen tissue sections.Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user
  • Target description: Dual specificity mitogen-activated protein kinase kinase 2 (UniProt: P36507; also known as EC:2.7.12.2, MAP kinase kinase 2, MAPKK 2, ERK activator kinase 2, MAPK/ERK kinase 2, MEK 2) is encoded by the MAP2K2 (also known as MEK2, MKK2, PRKMK2) gene (Gene ID: 5605) in human. MAP kinases are a group of evolutionarily conserved proline-directed protein serine/threonine kinases that are activated in response to a variety of extracellular stimuli and mediate signal transduction from the cell surface to the nucleus. They regulate several physiological and pathological cellular phenomena, including inflammation, apoptotic cell death, oncogenic transformation, tumor cell invasion, and metastasis. They are part of a three-tiered phospho-relay cascade consisting of MAP kinase, a MAP kinase kinase (MEK or MKK) and a MAP kinase kinase kinase (MEKK or MKKK). In this relay system MKKKs phosphorylate and activate MEKs, which in turn phosphorylate and activate MAPKs. MEKs are dual specificity enzymes that phosphorylate hydroxyl side chains of serine/threonine and tyrosine residues in their MAP kinase substrates. MEKs are activated by phosphorylation of serine or threonine residues in their activation loop. MEK2 plays a critical role in mitogen growth factor signal transduction. It phosphorylates a threonine and a tyrosine residue in a Thr-Glu-Tyr sequence located in MAPK1/ERK2 and MAPK2/ERK3. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
  • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
  • Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
