  • Target description: Mitogen-activated protein kinase 12 (UniProt: P53778; also known as EC:2.7.11.24, MAP kinase 12, MAPK 12, Extracellular signal-regulated kinase 6, ERK-6, Mitogen-activated protein kinase p38 gamma, MAP kinase p38 gamma, Stress-activated protein kinase 3) is encoded by the MAPK12 (also known as ERK6, SAPK3) gene (Gene ID: 6300) in human. p38g is a serine/threonine kinase that is an essential component of the MAP kinase signaling pathway. It plays an important role in the cascades of cellular responses evoked by extracellular stimuli such as proinflammatory cytokines and physical stress leading to direct activation of ELK1 and ATF2. It is highly expressed in skeletal muscle and heart during muscle differentiation and its knockdown is reported to impair cardiomyocyte formation. It also plays a role in blocking premature differentiation of myoblast and skeletal muscle stem cells. p38g is also required for the upregulation of PGC-1a in mitochondrial biogenesis in response to endurance exercise. It is activated by dual phosphorylation of Thr-183 and Tyr-185 by MKK3 and MKK6. It is shown to be overexpressed in highly metastatic breast cancer cell lines and its expression is preferentially associated with basal-like and metastatic phenotypes of breast tumor samples. Its expression is reported to be down-regulated by p38g and its interaction with the single PDZ domain of protein tyrosine phosphatase (PTPH1) leads to its dephosphorylation and inactivation. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
  • Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
