  • Specificity: Clone 1K3 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects cleaved form of Caspase-8. It targets an epitope within 23 amino acids from the cleavage site within the C-terminal half.
  • Immunogen: KLH-conjugated linear peptide corresponding to 23 amino acids from the cleavage site within the C-terminal half of mouse Caspase-8.

  • Target description: Caspase-8 (UniProt: O89110; also known as EC: 3.4.22.61, CASP-8, CASP8, MCH5, FLICE, MACH) is encoded by the Casp8 gene (Gene ID: 12370) in murine species. Caspase-8 is a cysteine protease that serves as a molecular switch for apoptosis, necroptosis, and pyroptosis, and is required to prevent tissue damage during embryonic development and adulthood. It is an initiator caspase that plays a major role in the extrinsic apoptotic signaling pathway via death receptors. It is expressed in wide variety of tissues and its highest expression has been reported in spleen, thymus, lung, liver, and kidney. Low levels of expression are observed in heart, brain, testis, and skeletal muscle. Caspase-8 is produced with pro-peptides (aa 1-218 and 377-387) that are cleaved under appropriate signals to produce a p20 (aa 219-376) and a p10 (aa 388-480) subunits that heterotetramerize to form active caspase-8 with p20 and two p10 subunits. Generation of the subunits requires association with the death-inducing signaling complex (DISC). Activated caspase 8 is subsequently released to the cytosol where it cleaves downstream effector caspases. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
  • Reconstitution: 300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
