  • Specificity: Clone 3B9 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects KEAP1. It targets an epitope within 17 amino acids from the Kelch repeat 2 within the C-terminal half.
  • Immunogen: KLH-conjugated linear peptide corresponding to 17 amino acids from the Kelch repeat 2 region within the C-terminal half of human Kelch-like ECH-associated protein 1 (KEAP1).
  • Application: Quality Control TestingEvaluated by Western Blotting in C6 cell lysate.Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detects KEAP1 in C6 cell lysate.   • Target description: Kelch-like ECH-associated protein 1 (UniProt: Q14145;  also known as Cytosolic inhibitor of Nrf2, INrf2, Kelch-like protein 19) is encoded by the KEAP1 (also known as INRF2, KIAA0132, KLHL10) gene (Gene ID: 9817) in human. KEAP1 is broadly expressed protein with highest levels observed in skeletal muscle. It serves as a substrate-specific adapter of a BCR (BTB-CUL3-RBX1) E3 ubiquitin ligase complex that regulates the response to oxidative stress by targeting Nrf2 for ubiquitination and degradation. It is reported to be a key regulator of the Nrf2, which transactivates the Antioxidant Response Element (ARE) and upregulates numerous proteins involved in antioxidant defense. Under basal conditions, KEAP1 targets Nrf2 for ubiquitination and proteolytic degradation.  However, in response to oxidative stress, different electrophile metabolites trigger non-enzymatic covalent modifications of highly reactive cysteine residues in KEAP1, leading to inactivate the ubiquitin ligase activity of the BCR(KEAP1) complex. This leads to nuclear accumulation of Nrf2 and expression of phase II detoxifying enzymes. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
  • Reconstitution: 300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
