  • Specificity: Clone 1F4 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Glycine amidinotransferase, mitochondria (GATM/AGAT). It targets an epitope within 22 amino acids from the N-terminal half.

  • Target description: Glycine amidinotransferase, mitochondrial (UniProt: P50440; also known as EC:2.1.4.1, L-arginine:glycine amidinotransferase, Transamidinase) is encoded by the GATM (also known as AGAT) gene (Gene ID: 2628) in human. GATM is a homodimeric protein that catalyzes the rate limiting step in the biosynthesis of guanidinoacetate, the immediate precursor of creatine. It displays an equilibrium between the monomeric and dimeric forms. It is synthesized with a mitochondrial transit peptide (aa 1-37), which is subsequently cleaved off to produce the active enzyme (aa 38-423). Its expression is observed in various tissues, with the highest expression observed in kidney. Its expression is elevated in the myocardium during cardiac failure. It may play a role in embryonic and central nervous system development and may also be involved in response to cardiac failure by elevating local creatine synthesis. Two isoforms of GATM have been described. Isoform 1 is localized in the mitochondrial inner membrane and isoform 2 is cytoplasmic. Mutations in GATM gene have been linked to cerebral creatine deficiency syndrome 3, an autosomal recessive disorder that is characterized by developmental delay, intellectual disability, and myopathy characterized by muscle weakness and atrophy. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
  • Reconstitution: 0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
