  • Target description: Granzyme B (UniProt: P10144; also known as EC:3.4.21.79, C11, CTLA-1, Cathepsin G-like 1, CTSGL1, Cytotoxic T-lymphocyte proteinase 2, Lymphocyte protease, Fragmentin-2, Granzyme-2, Human lymphocyte protein, HLP, SECT, T-cell serine protease 1-3E) is encoded by the GZMB (also known as CGL1, CSPB, CTLA1, GRB) gene (Gene ID: 3002) in human. Granzyme B, a serine protease of the peptidase S1 family, is synthesized with a signal peptide (aa 1-18) and a propeptide (aa 19-20) that are subsequently cleaved off to produce the active enzyme (aa 21-247). Granzyme B expression is regulated at both the transcriptional and translational levels and is influenced by many of the same factors that stimulate immune cell activation. Transcriptional activation of granzyme B within T lymphocytes involves activation of the T cell receptor and co-stimulation with cytokines. It is abundantly present in the cytosolic granules of cytotoxic T-cells and NK cells and activate caspase-independent pyroptosis when delivered into the target cells through the immunological synapse. Once delivered into the target cell, it acts by catalyzing the cleavage of gasdermin-E to release its pore-forming moiety. It is also linked to a caspase activation cascade that is responsible for apoptosis execution. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
  • Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
