- MRCK alpha (myotonic dystrophy kinase-related Cdc42-binding kinase alpha) is a member of the dystrophia myotonica protein kinase (DMPK) family that functions downstream of Cdc42 in actin cytoskeletal reorganization
- It is a serine/threonine kinase with multiple functional domains, including phorbol ester-responsive C1 domain
- Three independent coiled-coil domains and the N-terminal region preceding the kinase domain are responsible for intermolecular interactions leading to MRCK alpha multimerization
- The transautophosphorylation process is critical for regulation of MRCK alpha catalytic activities
- Binding of phorbol esters to MRCK releases its autoinhibition, allowing N-terminal dimerization and subsequent kinase activation.
