NB100-64329 is specific for human CD85a (leucocyte immunoglobulin-like receptor, subfamily B, member 3/ LILRB3), also known as ILT-5, expressed by monocytes, dendritic cells, macrophages, subpopulation of T lymphocytes, B cells and at high levels on plasma cells. CD85a is a 631 amino acid type I transmembrane glycoprotein, which contains four immunoreceptor tyrosine-based inhibition motif (ITIM) sequences within a long cytoplasmic tail. Phosphorylation of the tyrosine residues within ITIMs is known to enable the binding and activation of protein tyrosine phosphatases, which act as cell signalling modulators and inhibitors of cell activation.

NB100-64337 recognizes complement component 6 (C6), a 104 kDa member of the complement C6/C7/C8/C9 family present in blood serum. The factor I modules of C6 bind the C-terminus of the C5b alpha-chain, anchoring the molecule to the membrane; they also binds complement component 7. This enables the formation of the C5b-7 precursor complex. C5b-7 then functions as a receptor for components C8 and C9 which form the complement membrane attack complex (MAC). Deficiency of C6 results in susceptibility to bacterial infection. NB100-64337 recognizes MAC.

NB100-64339 recognizes 135 kDa complement component 3c (C3c), present in blood serum. C3c is generated over the course of complement activation, where convertase C4b2a (classical pathway) and convertase C3bBb (alternative pathway) cleave C3 to C3b and C3a. C3b is further degraded into iC3b and C3dg/C3d. iC3b is then slowly cleaved (possibly by factor I) to form C3c (beta chain + alpha' chain fragment 1 + alpha' chain fragment 2), C3dg and C3f. This antibody recognizes complement component 8 (C8), a 151 kDa member of the complement C6/C7/C8/C9 family, present in blood serum. C8 is a terminal component of the complement system, part of both the complement membrane attack complex (MAC), and important to MAC assembly. C8 binds to the C5b-7 complex, anchored to the membrane, creating C5b-8. C5b-8 binds C9 and catalyzes the polymerization of C9 molecules to form C5-b9 (MAC). C8 is thought to contain lipid binding sites, facilitating the insertion of MAC into the membrane. Defects in the alpha chain of C8 can result in complement C8 deficiency type I. Furthermore, C8 deficiencies can cause recurring bacterial infections, in particular from Neisseria meningitides.
