Test Specificity

Recognizes a protein of 36kDa, identified as cyclin D1. Cyclin D1, one of the key cell cycle regulators, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms. This antibody neutralizes the activity of cyclin D1 in vivo. About 60% of mantle cell lymphomas (MCL) contain a t(11; 14)(q13; q32) translocation resulting in over-expression of cyclin D1. This antibody is useful in identifying mantle cell lymphomas (cyclin D1 positive) from CLL/SLL and follicular lymphomas (cyclin D1 negative). Occasionally, hairy cell leukemia and plasma cell myeloma weakly express Cyclin D1.

This antibody recognizes a protein of 54kDa, which is identified as AMACR, also known as p504S. It is an enzyme that is involved in bile acid biosynthesis and oxidation of branched-chain fatty acids. AMACR is essential in lipid metabolism. It is expressed in cells of premalignant high-grade prostatic intraepithelial neoplasia (HGPIN) and prostate adenocarcinoma. The majority of the carcinoma cells show a distinct granular cytoplasmic staining reaction. AMACR is present at low or undetectable levels in glandular epithelial cells of normal prostate and benign prostatic hyperplasia. A spotty granular cytoplasmic staining is seen in a few cells of the benign glands. AMACR is expressed in normal liver (hepatocytes), kidney (tubular epithelial cells) and gall bladder (epithelial cells). Expression has also been found in lung (bronchial epithelial cells) and colon (colonic surface epithelium). AMACR expression can also be found in hepatocellular carcinoma and kidney carcinoma. Past studies have also shown that AMACR is expressed in various colon carcinomas (well, moderately and poorly differentiated) and over expressed in prostate carcinoma.

Test Specificity:
Actin is a major component of the cytoskeleton and is present in most cell types. It is highly specific to actin from smooth muscles. This monoclonal antibody does not stain cardiac or skeletal muscle; however, it does stain myofibroblasts and myoepithelial cells. This antibody could be used together with anti-muscle specific actin and myogenin in making a diagnosis of smooth muscle and skeletal muscle tumors. In most cases of rhabdomyosarcoma, this antibody yields negative results whereas anti-muscle specific actin and myogenin are positive. Leiomyosarcomas are positive only with anti-muscle specific actin and anti-smooth muscle actin and are negative with anti-myogenin.
