Test Specificity

This monoclonal antibody reacts with the beta-chain of HLA-DRB1 antigen, a member of MHC class II molecules. It does not cross react with HLA-DP and HLA-DQ. Its epitope is different from that of monoclonal antibody L243. HLA-DR is a heterodimeric cell surface glycoprotein comprised of a 36kDa alpha (heavy) chain and a 28kDa beta (light) chain. It is expressed on B-cells, activated T-cells, monocytes/macrophages, dendritic cells and other non-professional APCs. In conjunction with the CD3/TCR complex and CD4 molecules, HLA-DR is critical for efficient peptide presentation to CD4+ T cells. It is an excellent histiocytic marker in paraffin sections producing intense cytoplasmic staining. True histiocytic neoplasms are similarly positive. HLA-DR antigens also occur on a variety of epithelial cells and their corresponding neoplastic counterparts. Loss of HLA-DR expression is related to tumor microenvironment and predicts adverse outcome in diffuse large B-cell lymphoma.

Laminins are large hetero-trimeric, non-collagenous glycoproteins composed of alpha, beta, and gamma chains. This MAb reacts with laminin B2/1 chain of ∼210kDa and does not cross-react with other basement membrane components or fibronectin. Its specificity was established by immunoprecipitation and immunofluorescence of human skeletal muscle and kidney with laminin chain-specific MAbs. Epithelial sheets in vivo are separated from the mesenchymal elements of the stroma by a thin layer of a specialized type of extracellular matrix termed the basement membrane (BM). This structure consists of individual components, some of which are ubiquitous in BMs and some are not. The ubiquitous ones comprise laminin (LN), entactin/nidogen (EN), collagen type IV (CIV), and large heparan sulfate proteoglycan (HSPG), which interact specifically with each other to form a continuous and regular BM. Alterations of BM integrity, from local discontinuities up to complete loss, are described in many types of human and animal epithelial neoplasms. This MAb stains uniformly all human and murine basement membranes.

In Western blotting, it detects an antigen of 125kDa in human liver and 135kDa in tumors of histiocytic origin. Comparative study of this monoclonal antibody and a standard CD68 monoclonal antibody showed that their antigens are different. Its antigen in all macrophage types studied is located on the plasma membrane and within cytoplasmic structures including lysosomes. This monoclonal antibody shows a restricted reactivity to cells of the monocyte/macrophage system. It specifically reacts with blood monocytes and stains resident macrophages in a wide variety of human tissues. This monoclonal antibody does not stain antigen-presenting cells, e.g., Langerhans cells. Reportedly, its reactivity is restricted to histiocytes and macrophages.

It recognizes an intermediate filament protein (IFP) of 55kDa, which is identified as cytokeratin 7. This monoclonal antibody is highly specific to cytokeratin 7 and shows no cross-reaction with other IFPs. Cytokeratin 7 is a basic cytokeratin, which is found in most glandular and transitional epithelia but not in the stratified squamous epithelia. Keratin 7 is expressed in the epithelial cells of ovary, lung, and breast but not of colon, prostate, or gastrointestinal tract. This monoclonal antibody is highly useful in distinguishing ovarian carcinomas (keratin 7+) from colon carcinomas (keratin 7-).
