  • Target description: Succinate dehydrogenase [ubiquinone] cytochrome b small subunit, mitochondrial (CybS)(UniProt: O14521; also known as CII-4, QPs3, Succinate dehydrogenase complex subunit D, Succinate-ubiquinone oxidoreductase cytochrome b small subunit, Succinate-ubiquinone reductase membrane anchor subunit) is encoded by the SDHD (also known as SDH4 ) gene (Gene ID: 6392) in human. It is a multi-pass membrane protein and primarily localized in the inner motochondrian membrane. SDHD is one of four proteins that make up the tricarboxylic cycle enzyme succinate dehydrogenase (SDH). The specific role of SDH is to transfer electrons from succinate to ubiquinone, and the SDHD subunit is involved in anchoring the SDH complex to the inner mitochondrial membrane. SDHD is expressed in various tissues, with particularly high expression in tissues with high energy demands. Studies have shown that mutations in SDHD often leads to hereditary paragangliomas, usually benign tumors of the autonomic nervous system, suggesting that SDHD also plays a role as a tumorsuppressor gene. In one family with a nonsense mutation (R22X) in the SDHD gene, a loss of heterozygosity was found in the paragangliomas, and within these tumors the enzymatic activity of Complex II in the mitochondrial respiratory chain was completely abolished. Furthermore, high levels of angiogenic factors EPAS1 and VEGF was observed, which may stimulate tumor growth. Defective SDHD has been linked to Carney-Stratakis syndrome, a condition characterized by concurrent paraganglioma and gastric stromal sarcomas. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
  • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
  • Reconstitution: 300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
