Description

  • Differentiate Enterobacteriaceae according to their ability to ferment lactose, sucrose, and glucose, and to produce hydrogen sulphide using Thermo Scientific™ Oxoid™ Triple Sugar Iron Agar (Dehydrated)
  • The Triple Sugar Iron Agar is based on Kligler Iron Agar, additionally, it contains sucrose for the detection of sucrose fermenting species
  • The composite medium contains Peptone, 'Lab-Lemco' powder and yeast extracts to provide nitrogenous compounds and trace elements while sodium chloride maintains osmotic equilibrium
  • Lactose, sucrose and glucose are the fermentable carbohydrates
  • Sodium thiosulphate and ferric ions make the H2S indicator system while Phenol red is the pH indicator
  • Bacteria that ferment any of the three sugars in the medium will produce by-products
  • These by-products are usually acids, which will change the color of the phenol red to yellow
  • Position of the color change distinguishes the acid production associated with glucose fermentation from the acidic by-products of lactose or sucrose fermentation
  • Many bacteria that can ferment sugars in the anaerobic butt of the tube are enterobacteria
  • Typical reactions of organisms in Triple Sugar Iron Agar: Organism Slant Butt Gas H2S Escherichia coli ATCC® 8739 * A A + - Proteus hauseri ATCC® 13315* A A + + Pseudomonas aeruginosa ATCC® 9027 * ALK ALK - - Salmonella enteritidis ATCC® 13076 * ALK A + + ALK – Alkaline A – Acid Triple Sugar Iron Agar is recommended for the presumptive identification of colonies or sub-cultures from plating media such as Salmonella Shigella Agar (Modified) (CM0533), Bismuth Sulphite Agar (CM0201), Brilliant Green Agar (CM0263), MacConkey Agar No.3 (CM0115), or Desoxycholate Citrate Agar (Hynes) (CM0227)
