  • Holt-Harris and Teague 1 developed Eosin Methylene Blue Agar (EMB Agar)
  • The combination of dyes, lactose and sucrose incorporated in the medium provides a differential plating medium which helps to identify fermentative, gram-negative bacilli
  • Modified (HHT) EMB Agar w/Lactose, Sucrose is a solid medium used to differentiate gram-negative enteric bacilli

  • Lactose fermenting coliform and lactose nonfermenters are distinguished based on different colored colonies formed
  • Eosin and methylene blue dyes also act as selective agents to inhibit the growth of gram-positive organisms
  • The characteristic of certain coliforms to ferment lactose or sucrose can facilitate their identification and differentiation
  • Eosin dye combines with methylene-blue indicator to produce a color change when lactose or sucrose is fermented
  • Lactose fermenters such as Escherechia coli produce blue-black colonies with a green metallic sheen due to amide bonding of dye in an acidic environment
  • Other coliforms form mucoid, pink-brown colonies in less acidic conditions, whereas lactose nonfermenters Shigella and Salmonella form transparent, colorless or amber colored colonies
