• Biochem/physiol Actions: SMEK (suppressor of mek1) is a regulatory subunit of protein phosphatase 4 (PP4) complex. It consists of SMEK1 and SMEK2 subunits. SMEK is involved in histone deacetylation and silencing of the Wnt-signaling gene, brachyury, in embryonic stem cells (ESCs). It plays an important role in the recruitment of PP4 complex and histone deacetylase 1 (HDAC1) to the Tcf/Lef binding site of the brachyury promoter, inhibiting brachyury expression in ESCs. It associates with PP4c and represses transcription of Wnt-responsive development-related genes through histone deacetylation, maintaining the pluripotency of ESCs.
