• Biochem/physiol Actions: SH2D4A (SH2 domain containing 4A), a novel docking protein, belongs to the SH2 (Src homology 2) signalling protein family. It is ubiquitously expressed in various tissues and localizes in the cytoplasm. It is a single SH2 domain containing protein responsible for protein-protein interactions. It forms complex with the estrogen receptor α (ERα) to prevent the association of phospholipase C-γ (PLC-γ) with ERα. Thus, it has been reported that SH2D4A has the potential to inhibit cell proliferation by suppressing the ERα/PLC-γ/PKC (protein kinase C) signaling pathway. It may also play an important role in TCR (T cell receptor) signal transduction in T cells.
