Description

• General description: Merkel cells are neuroendocrine cells found in skin that have synaptic contacts with somatosensory afferents. These cells can turn malignant and form an aggressive form of skin cancer, which is known as Merkel cell carcinoma (MCC). A vast majority of MCC are caused by a polyomavirus known as Merkel cell polyomavirus (MCPyV or MCV). The MCPyV genome is reported to undergo clonal integration into the host cell chromosomes of MCC tumors and expresses small T antigen and truncated large T antigen. Full-length MCPyV large T-antigen is a 125 kDa nuclear protein, however, MCPyV T-antigens obtained from MCC have natural truncating mutations, which result in variably-sized, smaller proteins. Based on homology to other polyomaviruses, the MCPyV large and small T antigens are predicted to be oncogenic and contribute directly to the carcinogenesis of MCC. MCPyV large T antigen can serve as a specific marker for MCC.
• Specificity: Clone CM2B4 is highly specific for MCPyV large T and 57kT isoforms, but does not detect MCPyV small T antigen.
• Immunogen: KLH-conjugated linear peptide corresponding to RSRKPSSNASRGA sequence from the Merkel cell polyomavirus large T antigen exon 2 with a C-terminal cystiene.
• Application: Anti-MCPyV large T-antigen, clone CM2B4, Cat. No. MABF2044, is a mouse monoclonal antibody that detects MCPyV large T antigen and has been tested for use in Immunohistochemistry, Immunoprecipitation, and Western Blotting for the detection of Merkel cell polyomavirus.
• Quality: Evaluated by Western Blotting in MKL-1 cell lysates.Western Blotting Analysis: 1 µg/mL of this antibody detected MCPyV large T-antigen in 10 µg of MKL-1 cell lysates.
• Target description: 45 kDa observed for truncaed form. For intact form MW 125 kDaUncharacterized bands may be observed in some lysate(s).
