• Specificity: Clone 19M23 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects Tyrosine-protein kinase Tec. It targets an epitope within 19 amino acids from the N-terminal region.
• Immunogen: KLH-conjugated linear peptide corresponding to 19 amino acids from the N-terminal region of human Tyrosine protein kinase Tec.
• Application: Affinity Binding Assay: A representative lot of this antibody bound Tec peptide with a KD of 1.2 x 10-5 in an affinity binding assay. Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Tec in K562 cells.ELISA Analysis: A representative lot specifically detected Tec peptide in ELISA application, but not peptides from five other proteins.Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
• Target description: Tyrosine-protein kinase Tec (UniProt: P42680; also known as EC:2.7.10.2, Tec) is encoded by the TEC (also known as PSCTK4) gene (Gene ID: 7006) in human. The Tec family of protein kinases consists of five members, Tec, Btk, Itk/Emt/Tsk, Bmx and Txk/Rlk. They are abundantly expressed in hematopoietic cells and participate in the growth and differentiation of blood cells. Tec is a peripheral membrane protein that serves as a non-receptor tyrosine kinase that contributes to signaling from many receptors and participates as a signal transducer in multiple downstream pathways, including regulation of the actin cytoskeleton. It is activated by tyrosine phosphorylation by a wide range of cytokine stimulations. It regulates the development, function and differentiation of conventional T-cells and nonconventional NKT-cells and is required for TCR-dependent IL2 gene induction. Following B-cell or T-cell receptors activation by antigen, it is shown to translocate to the plasma membrane through its PH domain (aa 4-111). Tec is reported to serve as a central regulator of the noncanonical caspase-8 inflammasome that is involve in the assembly and activation of the caspase-8 inflammasome in response to fungal pathogens but is not involved in anti-bacterial immune responses. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
• Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
• Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
