  • Target description: Runt-related transcription factor 2 (UniProt: Q13950; also known as Acute myeloid leukemia 3 protein, Core-binding factor subunit alpha-1, CBF-alpha-1, Oncogene AML-3, Osteoblast-specific transcription factor 2, OSF-2, Polyomavirus enhancer-binding protein 2 alpha A subunit, PEA2-alpha A, PEBP2-alpha A, SL3-3 enhancer factor 1 alpha A subunit, SL3/AKV core-binding factor alpha A subunit, RUNX2) is encoded by the RUNX2 (also known as AML3, CBFA1, OSF2, PEBP2A) gene (Gene ID: 860) in human. RUNX2 is a master transcription factor that is specifically expressed in membranous and endochondral bones and is involved in osteoblastic differentiation and skeletal morphogenesis. Its expression in the bone and osteogenic front of a suture is crucial for cranial suture closure and membranous bone morphogenesis. It is a heterodimer of a and b subunits. The a subunit binds DNA as a monomer through its RUNT domain (aa 101-229). Its activity is shown to be precisely regulated by phosphorylation at multiple sites. Protein kinase C activated by FGF2 phosphorylates RUNX2 at serine 247 and ERK phosphorylates serine 301 in the PST domain that is reported to be critical for the enhancement of subsequent acetylation and the suppression of ubiquitination of the RUNX2 protein. Phosphorylation of serine 451 by CDK1 is reported to promotes endothelial cell proliferation. Mutations in RUNX2 gene have been linked to craniofacial malformations. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
  • Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
