  • Target description: Stromal antigen 2 (UniProt: Q8N3U4; also known as SCC3 homolog 2, Cohesin subunit SA-2, STAG2) is encoded by the STAG2 (also known as SA2) gene (Gene ID: 10735) in human. STAG2 is a component of a an evolutionarily conserved, multimeric protein complex, known as the cohesion complex, which is required for the cohesion of sister chromatids after DNA replication. In human somatic cells, cohesin consists of four core subunits: SMC1, SMC3, SCC1, and one of two related helical repeat proteins known as STAG1 or STAG2. This complex apparently forms a large proteinaceous ring within which sister chromatids can be trapped. The cohesin complex is also reported to play a role in spindle pole assembly during mitosis. At anaphase, the RAD21 subunit of cohesion is cleaved, which leads the dissociation of the complex from chromatin and allows sister chromatids to segregate. During prophase, most of cohesin complex is shown to dissociate from chromatin probably because of phosphorylation by PLK1. However, the complex remains intact at centromeres. In the cohesion complex, STAG2 gene exhibits high mutation rate with about 90% mutations resulting in the introduction of premature codons that leads to loss of protein function. STAG2 mutations are known to cause Mullegama-Klein-Martinez syndrome, a neurodevelopmental disorder leading to intellectual deficiency, hearing loss, developmental delay, and congenital heart defect.   • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
  • Reconstitution: 300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
