Description

  • General description: We are committed to bringing you greener alternative products, which adhere to one or more of the 12 Principles of Green Chemistry. This antibody is preservative-free, produced without the harm or sacrifice of animals and exceptionally stable to allow for ambient shipping and storage if needed, and thus aligns with "Waste Prevention", "Designing Safer Chemicals" and "Design for Energy Efficiency". Click here for more information.
  • Specificity: Clone 5210-87-133 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Human APOBEC3B. It targets an epitope within 28 amino acids from the C-terminal region. May cross-react with human A3A and A3G.
  • Immunogen: KLH-conjugated linear peptide corresponding to 28 amino acids from the C-terminal region of human DNA dC->dU-editing enzyme APOBEC-3B (APOBEC3B).
  • Application: Quality Control TestingEvaluated by Western Blotting in U2OS cell lysate.Western Blotting Analysis: A 1:1,000 dilution of this antibody detected APOBEC3B in U2OS cell lysate.Tested ApplicationsWestern Blotting Analysis: A 1:1,000 dilution from a representative lot detected APOBEC3B in MCF7 and THP-1 cell lysates.Immunohistochemistry (Paraffin) Analysis: A 1:25 dilution from a representative lot detected APOBEC3B in Human breast cancer, Human cervical cancer and Human bladder cancer tissue sections.Enzyme Immunoassay (ELISA) Analysis: A representative lot detected APOBEC3B in ELISA applications (Brown, W., et. al. (2019). Antibodies (Basel). 8(3):47).Immunocytochemistry Analysis: A 1:25 dilution from a representative lot detected APOBEC3B in U2OS cells.Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
  • Target description: DNA dC->dU-editing enzyme APOBEC-3B (UniProt: Q9UH17; also known as EC:3.5.4.38, A3B, Phorbolin-1-related protein, Phorbolin-2/3) is encoded by the APOBEC3B gene (Gene ID: 9582) in human. APOBEC3B is a member of the cytidine deaminase gene family. It contains a conserved zinc-binding domain and a C-terminal catalytic domain that is responsible for its deaminase activity. Its pseudo-catalytic N-terminal domain can bind to nucleic acids but does not have deaminase activity. APOBEC3B can cause C-to-U mutations in single-stranded DNA molecules, subsequently inducing hypermutation in the viral genome. APOBEC3B primarily targets retroviruses and retrotransposons. APOBEC3B is expressed in various tissues, including the reproductive organs, lymphoid tissues, and epithelial cells of the respiratory and gastrointestinal tracts. Its expression is up-regulated by - interferon. APOBEC3B has been implicated in the development of several diseases. It has been associated with carcinogenesis, particularly in breast cancer. Elevated expression of APOBEC3B in cancer cells can lead to increased mutagenesis and genomic instability. APOBEC3B-induced mutations contribute to the evolution of tumor genomes and the development of drug resistance. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. (Ref.: Brown, WL., et al. (2019). Antibodies (Basel). 8(3); 47, Maiti, A., et al. (2021). Curr. Opin. Struct. Biol. 67; 195-204)
  • Physical form: Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
  • Reconstitution: 300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
  • Storage and Stability: Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
