Description

  • Description: The NK92.39.1 monoclonal antibody reacts with human CD96 also known as TACTILE (T cells-activated increased late expression)
  • CD96 is a type I membrane protein of the Immunoglobulin Superfamily
  • CD96 promotes NK cell adhesion to cells expressing the polio virus receptor (CD155)
  • Therefore CD96 is an additional receptor for CD155
  • After binding of PVR to CD96, the PVR can be transferred to the target cell and CD96 can be internalized with bound PVR
  • This internalization may lead to cell death of the NK cell
  • This phenomenon may be significant in tumor cells which express high levels of PVR
  • Furthermore expression of CD96 has been found in the majority of CD34(+)CD38(-) Acute Myeloid Leukemic cells
  • The NK92.39.1 antibody can block binding of soluble CD155 to NK cell lines
  • Applications Reported: This NK92.39 antibody has been reported for use in flow cytometric analysis
  • Applications Tested: This NK92.39 antibody has been pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells
  • This can be used at 5 μL (0.5 μg) per test
  • A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL
  • Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test
  • Excitation: 488-561 nm; Emission: 578 nm; Laser: Blue Laser, Green Laser, Yellow-Green Laser

  • CD96 is a cell adhesion receptor also known as TACTILE
  • A member of the Ig superfamily, CD96 is a single pass transmembrane protein consisting of three extracellular Ig-like domains and a proline/serine/threonine rich region
  • CD96 is expressed on resident monocytes and T cells at all developmental stages (including gamma-delta T cells, thymocytes, and NK cells)
  • In the mouse, this receptor is not expressed on B cells
  • Unlike in humans, mouse CD96 does not exist as alternative splice variants
  • Reports indicate that CD96 binds CD155/poliovirus receptor and Nectin-1 to mediate NK cell adhesion to target cells and cytotoxicity.
